Introduction
In this study, we use Synthace to design and execute a flexible automated PicoGreen™ assay. PicoGreen™ assays are high sensitivity fluorometric protocols for measuring DNA concentration that have a wide range of applications, including tracking bioprocesses, quantifying viral vector concentrations, and testing patient samples.
The specific PicoGreen™ DNA binding dye overcomes the limitations of traditional absorbance-based DNA quantification methods, which are susceptible to the presence of proteins, RNA, and single-stranded DNA in the original sample leading to measurement inaccuracies.
However, manual PicoGreen™ assays have some limitations. Firstly, variations in methodology, reagent preparation, and timing of execution lead to variable outcomes, resulting in poor data reproducibility. Secondly, manual execution is laborious and time consuming, preventing scientists from focussing on more valuable tasks.
Key Findings
- We performed an automated PicoGreen™ assay using our cloud-based software platform Synthace
- Synthace enabled flexible design, planning, and optimization of the assay, followed by its seamless execution on a Tecan Freedom EVO® 100 liquid handling robot
- Synthace achieved results comparable to manual execution but with higher dilution accuracy (R²>0.999) and lower variability between technical replicates (%CVs < 1% in Synthace runs vs. > 2% in manual runs)
- Compared to manual execution, Synthace reduced hands-on time by 45% per plate (55min to 30min)

Compared to manual PicoGreen™ assays, automated execution using Synthace streamlined the process, reducing the number of steps required to carry out the assay and minimizing researcher hands-on time by 45% per plate: from 55 min/plate in manual execution to 30 min/plate in automated execution with Synthace.

In this study, we showed that Synthace enables scientists to accelerate the design, planning, optimization, execution of flexible automated workflows for plate based assays, such as DNA quantification using PicoGreen™, without the need for automation or coding expertise.
